Effects of mescaline and mechanism of action

Mescaline is obtained naturally from the Peyote and San Pedro cactuses; it is classified as a chemical entity within the class of phenethylamines (the psychoactive or “psychostimulant” portion of these drugs), which includes a number of stimulants, including dopamine, norepinephrine, and epinephrine, plus numerous others, including MDMA.

A typical mescaline dose ranges from 300 to 500 mg, with common psychedelic effects including euphoria, time distortion, perceptual changes, visual hallucinations, and ego death, especially at high doses. Mescaline has a delayed onset of action compared to magic mushrooms, possibly due to slow brain penetration. It is also more likely to induce pronounced nausea and vomiting, which is often seen as a cleansing process in traditional rituals.

Although mescaline binds with the serotonin 5-HT2A receptor (all psychedelics are known to bind with this receptor), it affects both the dopamine and norepinephrine pathways. Unlike mescaline, psilocin does not significantly bind to dopamine or adrenaline receptors. This receptor selectivity is why psilocybin tends to produce more consistent "mystical-type" experiences and clearer emotional shifts without the heavy physical "push" associated with phenethylamines.

Potency, dosage, and duration

Mescaline and psilocybin are very different in terms of their overall effect on the body. While mescaline is less potent by weight, it generally takes high doses (200 - 500 milligrams) for an individual to experience a full-on psychedelic experience, due to differences in the amount of active ingredients in the raw plant form (cacti) used to create mescaline.

Because of the differences in how mescaline is typically consumed, there will be a great deal of variation in how each individual can dose and prepare the psychedelic they are taking. In addition to the differences in active ingredient concentration, the duration of the effects of mescaline versus psilocybin also shows considerable differences.

A mescaline journey is considered an endurance event. Because mescaline usually takes 12-16 hours to completely dissipate, individuals who choose this substance need to devote themselves for 12 + hours, not just the time spent ingesting but also the time required after taking this psychedelic to come down and learn to re-assimilate themselves into everyday life.

Psilocybin's therapeutic window is much shorter than mescaline. Whereas the therapeutic window for mescaline can range between 10-12 hours (or longer); the therapeutic window for psilocybin is approximately four to six hours.

The shorter length of a journey involving psilocybin allows for adequate time to have a full experience, as well as a reasonable amount of time for grounding and initial integration, all within the same day.

By contrast, psilocybin provides a very inward and immersive experience. The chance of having a “mystical-type” experience is greater with psilocybin than with mescaline; that is, the experience has the potential to bring about states of consciousness in which the ego has disappeared and a person feels an overwhelming sense of connection to the universe.

Visually, psilocybin also has a tendency to be more inward-looking than mescaline and often includes many of the archetypal images describing deep emotional landscapes.
